Investigating how autoimmune T cells are generated

We are interested in how autoimmune T cells are generated. We study in situ T cell activation in lymph nodes from a variety of locations in both mice and patients with inflammatory disease. We are generating live lymph node slice culture systems, and we use immunofluorescence and histochemical staining as well as flow cytometry and functional assays.

Neutrophilic infiltration of lymph nodes increases throughout the development of inflammatory disease. Inguinal lymph nodes showing pink neutrophil migration (staining for the granule peptide cathelicidin) during time course of the mouse model of Multiple Sclerosis, EAE. We are interested in using slice cultures of these nodes to understand where the neutrophils move to, and what impact they have on the development of T cell immunity in the nodes.
